basebuild-v4 and SDHC cards

Hi, Just waiting on my Zgemma h2s to arrive and getting a few things ready.

I have a good few 16GB SDHC cards and was wonder in I could use one of these with a Card Reader for the SDHC Cards?
For first flashing with the basebuild-v4 (in front of box) and then use a different one in the back of the box for the rest of the setup?
